The Dawn of War series of video games is probably the best place to start with them. The universe generally favours brute violence and massive wars as the solution to most problems, though. There are also psykers, prophecies and an immortal emperor living for thousands of years with a plan to save the human race but only at a heavy cost. The Warhammer 40,000 universe is somewhat inspired by Dune (as well as Aliens and Starship Troopers), and although it does have aliens the main threat comes from 'tainted' or corrupt humans.

The 'humans are the only intelligent race' thing is particularly rare in video games, as they need more different factions for gameplay purposes. Not really, although there's some in the same general wheelhouse.
